研究目的
To promote the discovery of biomarkers of bacterial infection by characterizing gastric cells infection by diverse Helicobacter pylori strains through Fourier-transform infrared spectroscopy.
研究成果
FTIR spectroscopy enabled the discrimination of AGS cells incubated with H. pylori from those not incubated, and further distinguished between different H. pylori genotypes and the gastric diseases of the patients from which the bacteria were isolated. This method offers a rapid, economic, and high-throughput platform for understanding the complex infection process and developing new diagnostic methods.
研究不足
The study is limited by the specific genotypes of H. pylori strains used and the focus on AGS cells, which may not fully represent the diversity of gastric cell responses to H. pylori infection.
1:Experimental Design and Method Selection:
FTIR spectroscopy was used to acquire spectra from adenocarcinoma gastric cells incubated with H. pylori strains of different genotypes.
2:Sample Selection and Data Sources:
AGS cells were incubated with H. pylori strains presenting different genotypes concerning the virulent factors CagA and VacA.
3:List of Experimental Equipment and Materials:
FTIR spectrometer (Vertex 70, Bruker), CO2 incubator (Blinder CB150), Axiovert 40 CFL microscope (Carl Zeiss), and 96-wells Si plate.
4:Experimental Procedures and Operational Workflow:
AGS cells were incubated with H. pylori strains, spectra were acquired, pre-processed, and analyzed.
5:Data Analysis Methods:
Principal component analysis (PCA) and statistical hypothesis tests were used to evaluate the absorbance ratios.
