To investigate the role of broadband and monochromatic photopic negative response (PhNR) of the full-field flash electroretinogram (ERG) in the evaluation of ganglion cell damage in primary open-angle glaucoma (POAG) subjects.

Red-on-blue PhNR is a useful additional diagnostic tool for detecting glaucomatous neural damage, showing better sensitivity and correlation with structural and functional tests compared to other stimuli. It can complement existing clinical assessments like visual field tests and OCT.

The study was non-randomized and had a relatively small sample size. Longitudinal studies with larger samples are needed to evaluate the role of PhNR in various stages of glaucoma and response to therapy. The study did not include ocular hypertensives.
1:Experimental Design and Method Selection:
A prospective, cross-sectional, case-control study was conducted. ERG was recorded using broadband and monochromatic stimuli to compare PhNR responses. Statistical analyses included ANOVA, MANOVA, Pearson correlation, and ROC curves.
2:Sample Selection and Data Sources:
25 POAG patients and 50 age-matched normal subjects were recruited from a tertiary eye care center in South India. Inclusion criteria involved ophthalmic evaluations and specific visual field and OCT measurements.

ERG recording system (VERIS Ganzfeld), electrodes (Burian-Allen contact lens electrodes), tonometer (Zeiss AT 030 Applanation Tonometer), gonioscope (Posner-type 4 mirror), OCT (Cirrus HD), eye drops (tropicamide, hypromellose), and statistical software (SPSS, Excel).

Participants underwent dark adaptation, electrode placement, and ERG recording with different stimulus conditions. PhNR amplitude and implicit times were measured.
