  • A dual-responsive fluorescent probe for detection of fluoride ion and hydrazine based on test strips

    摘要: Hydrazine (N2H4) and fluoride ion (F-) are regarded as environmental pollutants and potential carcinogens. A dual-functional fluorescent probe (probe 1) was developed for both F- and N2H4 with high selectivity and sensitivity. 1 was based on nucleophilic aromatic substitution reaction for N2H4 detection and selective cleavage of 4-nitrobenzenesulphonyl group for the determination of F-. The limits of detection of probe for F- and N2H4 were 77.82 nM and 29.34 nM, respectively, which are far below the threshold limit value (TLV) of United States Environmental Protection Agency (EPA). The home-made test strips of 1 provided the positive tool for F- and gaseous N2H4 in different system. And the confocal fluorescence images indicated that 1 can quantitatively detect N2H4 in living PC12 cells. Promisingly, 1 has great prospects for N2H4 imaging and determining in living system.

  • A series of BODIPY-based probes for the detection of cysteine and homocysteine in living cells

    摘要: Biothiols, such as glutathione (GSH), homocysteine (Hcy) and cysteine (Cys), are important biomarkers and play crucial roles in many physiological processes. Thus, the detection of biothiols is highly important for early diagnosis of diseases and evaluation of disease progression. Herein, new types of BODIPY-based fluorescent probes (probe 1, probe 2 and probe 3) capable of cysteine (Cys)/homocysteine (Hcy) sensing with high selectivity over other amino acids were developed. In addition, we further studied the influence of different electronegativity substituents on these probes to sensing Cys/Hcy. Ultimately, we concluded that the electron withdrawing group on probe 1 can accelerate the probe response to Cys/Hcy, and probe 1 was successfully applied for selective imaging Cys/Hcy in living cells.

  • A new strategy for utilizing activated CH-group to construct a FRET platform for ratiometric sensing of cyanides

    摘要: A fluorescent and colorimetric cyanide sensor (4-Br) based on the activated C-H group to construct a FRET platform has been described for the first time, along with demonstration of selective and reversible detection of cyanides through it. The sensing mechanism of 4-Br is an integration of ICT and FRET mechanisms, based on the deprotonation of the activated C-H group. Importantly, it is suitable for fluoresence imaging of cyanides in living cell.

  • Time-Resolved Structured Illumination Microscopy for Phase Separation Dynamics of Water and 2-Butoxyethanol Mixtures: Interpretation of “Early Stage” Involving Micelle-Like Structures

    摘要: Phase separation dynamics of a water/2-butoxyethanol (2BE) mixture was studied with newly developed time-resolved structured illumination microscopy (SIM). Interestingly, an employed hydrophobic fluorescent probe for SIM showed spectral shifts up to 500 ns after a laser-induced temperature jump, which suggests 2BE micellar-like aggregates become more hydrophobic at the initial stage of phase separation. This hydrophobic environment in 2BE aggregates, probably due to the ejection of water molecules, continued up to at least 10 μs. Time-resolved SIM and previously-reported light scattering data clearly showed that the size of a periodic structure remained constant (ca. 300 nm) from 3 to 10 μs, and then the growth of periodic structures having the self-similarity started. We think that the former and the latter processes correspond to “early stage” (concentration growth) and “late stage” (size growth), respectively, in phase separation dynamics. Here we suggest that, in the early stage, the entity to bear 2BE phase be water-poor 2BE aggregates, and the number density of these aggregates would simply increase in time.

  • Iridium(III) complex-based fluorescent probe for detection of thiophenols and its application in water samples

    摘要: In consideration of the nucleophilic capacity of thiophenols, which could cleave sulfonamide under neutral conditions, a cyclometalated iridium(III) complex (probe 1) comprised 2,4-dinitrobenzenesulfonamide (DNBS) as the recognition unit was designed and synthesized to high selectivity and sensitivity detect thiophenols successfully based on internal charge transfer (ICT) mechanism. Upon addition with thiophenols to 20% DMSO/PBS solution of nearly non-fluorescent probe 1 (10 μM, pH = 7.4), fluorescent product (complex 2) was formed after cleavage of DNBS group and a significant emission enhancement at 470 nm was observed. In addition, it could be successfully applied to detect thiophenol in water samples with a high recovery, confirming its practical value in environment science.

  • Biothiol detection by “ON-OFF-ON” fluorescence probe based on anthracene derivative

    摘要: Two thiols fluorescent probes of copper complex based on anthracene derivative was designed. Two compounds (1 and 2) were obtained by the reaction of 9-furaldehyde with benzenesulfonamide and p-nitrobenzenesulfonamide. Then, these compounds were combined with copper ion and two copper complexes (compound 1-Cu2+ and compound 2-Cu2+) formed. Due to the paramagnetic effect of copper caused fluorescence quenching, two compounds showed strong fluorescence while compound-Cu2+ almost had no fluorescence. With the additions of cysteine (Cys) and glutathione (GSH), the fluorescence intensities recovered again. While the additions of other amino acids almost did not induce any change. UV-Vis, fluorescence spectra and Circular Dichroism suggested that two copper complexes showed better selectivity and sensitivity to small molecule biothiols among amino acids tested. In addition, cytotoxicity experiments showed that two fluorescence probes have potential application value for biological thiols detection in live cells.

  • A novel colorimetric and ratiometric fluorescent probe for selective detection of bisulfite in real samples and living cells

    摘要: An abnormal level of bisulfite can induce toxicological effects associated with lung cancer, cardiovascular and neurological disorders. Therefore, it is of significance to develop an effective fluorescent probe to detect bisulfite in living cells. Herein, a novel fluorescent probe QPCT, based on a 1,4-addition mechanism, was constructed for the colorimetric and ratiometric detection of bisulfite. QPCT displayed high selectivity and anti-interference ability to bisulfite over other anions and biothiols. The probe renders a sensitive ratiometric response to bisulfite with a remarkable fluorescence blue shift from 590 to 537 nm and the fluorescence ratio was linear with bisulfite concentration over the range of 0-120 μM. More importantly, QPCT has been successfully applied to detect bisulfite in sugars and living A549 cells, which indicated that QPCT had a great capability for monitoring bisulfite in complex systems.

  • A smart approach toward rhodamine spiro-ring derivatives sensing platform for lysosome-targetable imaging applications

    摘要: As acidic organelles (pH 4.0-5.5) in living cells, lysosomes play key roles in numerous cellular biological processes, and associate with diverse diseases. Small molecular probes based fluorescence imaging techniques capable of providing information on target analytes in lysosomes are highly appreciated. Herein, for the first time we propose a new strategy of constructing pH-independent rhodamine based spiro-ring platform for lysosomal imaging applications. As a proof of concept, by incorporating lysosome specific moiety morpholine, a thio-lactone rhodamine analogue HN4MS for HOCl with high sensitivity, high selectivity and fast response at lysosomal pH value was developed. Moreover, possessing ability of imaging in lysosome organelles, the new probe was successfully applied to visualize exogenous and endogenous HOCl in living cells and tissues, which verified the present avenue for expansion of organelle targetable imaging tools.

  • Imaging of endogenous hydrogen peroxide during the process of cell mitosis and mouse brain development with a near-infrared ratiometric fluorescent probe

    摘要: Hydrogen peroxide (H2O2) is emerging as a new second messenger, which plays vital roles in intracellular signaling, thereby triggering physiological variations in terms of proliferation, differentiation, and migration. As known, cell mitosis has closely association to the intracellular level of H2O2, which contribute to the significant effects on the brain development, especially during the critical period of immaturity. Unfortunately, imaging H2O2 in mammalian brain is still challenging. Herein, to further investigate the biological roles of endogenous H2O2 in cells mitosis, we develop a near-infrared ratiometric fluorescent probe Cy-PFS for specifically imaging of endogenous H2O2 in cells and in vivo. Employing the probe Cy-PFS, we examine the critical effects of endogenous H2O2 on cells proliferation in live hippocampal neurons cells, our results provide strong evidences for H2O2 signaling in cell mitosis through growth factor signaling. Furthermore, we successfully demonstrate the close association of endogenous H2O2 level changes with the brain development at various stages. We envision that this present probe has potential as a promising useful chemical imaging tool for exploring the roles of H2O2 in cell mitosis.

  • Phosphorus and chlorine co-doped carbon dots with strong photoluminescence as a fluorescent probe for ferric ions

    摘要: Usually, carbon dots (CDs) display a relatively weak fluorescence quantum yield (QY). In order to obtain brighter CDs, phosphorus and chlorine co-doped CDs (P,Cl-CDs) were prepared via hydrothermal treatment of maltose in the presence of phosphoric and hydrochloric acids. The new CDs are highly monodispersed in water solution, have high fractions of P (14.4 atomic%) and Cl (8.9 atomic%), and exhibited yellow fluorescence with a QY of 15%. This is higher than that of monoatomic doped CDs (8.7 and 9.3% for P-CDs and Cl-CDs, respectively). The P,Cl-CDs are highly photostable, and fluorescence is strongly (statically) quenched by Fe(III). Fluorescence decreases with increasing concentration of Fe(III) in the range from 0.1–8.0 μmol?L-1, with a 60 nmol?L-1 detection limit. The doped CDs are shown to be a viable nanoprobe for the fluorometric determination of Fe(III) in spiked serum and water samples.
