The combined hydrogen bonding and π-π stacking interactions in PEG-PAI4 enable a large red-shifted emission and self-assembly into nanovesicles, effectively suppressing fluorescence quenching in water. The material exhibits excellent biostability, photostability, and thermoresponsiveness, making it a promising candidate for cell imaging. Future work should focus on optimization for in vivo applications.

1:Experimental Design and Method Selection:
The study involved synthesizing PEGylated poly(amide-imide) (PEG-PAI4) via polycondensation of p-xylylenediamine, thiolactone-maleimide monomer, and poly(ethylene glycol) methyl ether methacrylate, utilizing hydrogen bonding and π-π stacking interactions for self-assembly into nanovesicles. Theoretical models like MM2 energy minimization were used for structure optimization.
2:Sample Selection and Data Sources:
Polymers such as PEG-PAI4, PEG-PAI1, and PEG-co-PSUC1 were synthesized and characterized. Cell lines (BEL-7402, 4T1, Hela) were used for biocompatibility and imaging studies.

Materials included DL-homocysteine thiolactone hydrochloride, 2-bromoisobutyryl bromide, maleic anhydride, sodium L-ascorbate, copper sulfate pentahydrate, sodium azide, diamines, PEG derivatives, AIBN, solvents, and dyes like rhodamine 6G.

4. Experimental Procedures and Operational Workflow: Synthesis involved dissolving monomers in DMSO, adding diamine, stirring, and purifying by precipitation. Self-assembly was done via nanoprecipitation: dissolving polymer in DMSO, adding to water under stirring, dialyzing, and concentrating. Cell imaging involved incubating cells with PEG-PAI4, washing, fixing, and imaging with CLSM. Biocompatibility was assessed using CCK-8 assay.

Data were analyzed using NMR spectroscopy, fluorescence and UV-vis spectroscopy, GPC for molecular weight, DLS and TEM for size measurement, FTIR for hydrogen bonding, and statistical methods for cell viability assays.
